Course Objectives
Capillary Electrophoresis (CE) is a rapidly developing, comparatively new separation technique. CE has very wide applications from the smallest Li ion to DNA. Small sample consumption, fast analysis time, high separation efficiency are distinct advantages of this technique. Numerous original publications have been reported on capillary electrophoretic applications on food, pharmaceutical, environmental, clinic, and forensic science areas. Recently, CE has been used as the main separation technique in genomic and proteomic researches. This lecture aims to inform the students in broad spectrum (chemistry, food engineering, environmental engineering, and molecular biology) about CE.
Course Description
Principles of CE / Factors affecting the performance of CE separations: Ion mobility, solution pH, ionic strength, use of additives in the background electrolyte, electroosmos, electroosmotic flow profile, voltage source, field strength, electrophoretic mobility, conductivity of solutions, electrodes. / Capillary columns: Coating techniques / The efficiency of CE: Migration times, zone broadening, plate numbers, resolution / Injection methods: Hydrodynamic injection, electrokinetic injection, influence of sample matrix/detection methods: on-column detection, UV detection, florescence detection, mass, indirect detection. / Quantitative aspects of CE analysis / Capillary zone electrophoresis / Micellar electrokinetic chromatography / Capillary electrochromatography /Capillary gel electrophoresis / Isoelectric focusing / CE in entangled polymer solutions / Application of CE: Drugs and related compounds, food samples, clinical samples, forensic science, biopolymers (proteins, DNA, carbohydrates), chiral separations, synthetic polymers, separation of small molecules
